Drosophila melanogaster kā modeļorganisms pētījumos par sēņu Lentinula edodes ekstraktu ietekmi uz novecošanas procesiem

Darbā pētīta šitakē sēnes micēlija ekstrakta ietekme uz augļu mušas Normal un Oregon līniju dzīves ciklu un virgīnu imago dzīves ilgumu un lokomotoro aktivitāti atkarībā no mušu vecuma. D. melanogaster barošanai ar ekstraktu izmantotas divas pieejas: i) piedeva iemaisīta barotnē, un ar to baroti indivīdi pre-imago stadijā, vai ii) piedeva pievienota rauga suspensijai, uznesta uz barotnes virsmas, un ar to baroti indivīdi imago stadijā, sākot no 24h vecuma - visu dzīves laiku. Ņemot vērā ar etanolu precipitējamās frakcijas sausnas daudzumu micēlija ekstraktā, sastādītas četras koncentrācijas grupas: 0,000% (kontrole), 0,015%, 0,030% un 0,100% sausna / barotnes vai rauga suspensijas tilpumā. Darba gaitā novērtēti tādi parametri kā: i) kūniņu un izšķīlušos imago skaits, dzīves cikla garums, ieskaitot kāpura stadijas attīstības ilgumu, kūniņu un mušu izdzīvotība, ii) imago letalitāte, atkarība no vecuma, un iii) dažāda vecuma indivīdu negatīvais ģeotropisms un spontānā aktivitāte. In this work was investigated shiitake mushroom mycelium extract effect on fruit flies Normal and Oregon life cycle and virgin imago life span and locomotor activity depending on fly age.
To feed D. melanogaster with the extract was used two approaches: i) an additive was mixed into the medium and the individuals were fed under pre-imago stage, or ii) an additive was added to the yeast suspension, applied on the surface of the medium, with what the imago stage individuals vere fed, ranging from 24 hours of age during lifetime. Considering the precipitated with ethanol fraction dry amount in mycelium extract, made up four groups of concentrations: 0.000% (control), 0.015%, 0.030% and 0.100% dry matter / in media or in yeast suspension volume. In this work were measured parameters such as: i) larvae and hatched imago number, life cycle length, including the larval stage developing length, larvae and fly survival, ii) the imago lethality, depending on age, and iii) negative geotropisms
