Bakteriofāga Enc34 replikācijas moduļa proteīna ORF6 struktūras noteikšana un analīze

Abstract

Pašlaik genoma datubāzēs ir deponētas vairāk nekā 1200 bakteriofāgu (fāgu) genomu sekvences. Vairums fāgu proteīnu tiek apzīmēti kā konservatīvi vai hipotētiski, un to struktūra un funkcijas pašlaik nav zināmas. Darba mērķis bija noteikt fāga Enc34 proteīna ORF6 terciāro struktūru un prognozēt tā funkciju. Pētījumā sekmīgi tika veikta natīva un ar selenometionīnu iezīmēta ORF6 proteīna ekspresija, attīrīšana un kristalizācija. Ar rentgenstaru kristalogrāfiju noteiktajai ORF6 struktūrai ir ssDNS saistošiem proteīniem (SSB) raksturīga uzbūve ar oligonukleotīdu/oligosaharīdu saistīšanas folda domēnu un nestrukturētu C-terminālo segmentu. Par ORF6 piederību SSB saimei liecina arī strukturālā homoloģija ar fāga T7 SSB.

At the moment, more than 1200 bacteriophage (phage) genomic sequences have been deposited in genome databases. The majority of phage proteins are annotated as conservative or hypothetical with an unknown structure and function. The main objective of this work was to determine the tertiary structure of phage Enc34-encoded protein ORF6 and to propose its function. In the study, expression, purification and crystallization of native and selenomethionine-labelled ORF6 protein were accomplished. The structure of the ORF6 protein, determined by X-ray crystallography, is characteristic for ssDNA-binding proteins (SSB) with an oligonucleotide/oligosaccharide-binding fold domain and a disordered C-terminal segment. Structural homology with an SSB from phage T7 further confirms that ORF6 is a member of the SSB family.
