dc.description.abstract | Neskatoties uz to, ka ir izstrādāta droša un efektīva hepatīta B vakcīna, kā arī vesela rinda pret hepatīta B vīrusu (HBV) vērstu ķīmisku līdzekļu, pasaulē ir >350 miljonu hronisko HBV nēsātāju, no kuriem 1 miljons mirst katru gadu. Galvenais jaunu terapijas veidu izstrādāšanas kavēklis ir efektīvas HBV infekciju un replikāciju nodrošinošas in vitro modeļsistēmas trūkums. Pateicoties revolucionāram atklājumam par NTCP (sodium taurocholate cotransporting peptide) lomu HBV infekcijas procesā pašlaik tiek aktīvi izstrādātās jaunās HBV in vitro infekcijas modeļsistēmas.
Mūsu darbā NTCP receptoru ekspresējošo šūnu iegūšanai tika izmantota efektīva Semliki Meža vīrusa (SFV) replikona ekspresijas sistēma. NTCP proteīna ērtai detekcijai tā C-galam tika pievienots zaļais fluorescentais proteīns (EGFP). Pielietojot gēnu inženierijas metodes tika izveidotas divas plazmīdu konstrukcijas pEF4 NTCP-EGFP un pSFV1/NTCP-EGFP, kas satur NTCP-EGFP gēnu. Lai pētītu NTCP ekspresiju, tā lokalizāciju un funkcionalitāti pēc attiecīgo konstrukciju ievadīšanās BHK-21 un HepG2 šūnās, tās analizēja ar imūnfluorescences metožu palīdzību. Parādīta efektīva NTCP-EGFP gēna ekspresija un NTCP-EGFP proteīna lokalizācija šūnu plazmas membrānā un to spēja saistīt HBV preS1 peptīdu. | en_US |
dc.description.abstract | Despite of the fact that is designed safe and effective hepatitis B vaccine, as well as a long line against hepatitis B virus (HBV) oriented chemical agents, in the world there are >350 million chronic carriers of the HBV, of which 1 million die each year. The main impediments of the developpement of a new therapies are an effective HBV infection and replication in the vitro model systems-providing drawback. Thanks to the NTCP (sodium taurocholate cotransporting peptide) revolutionary discovery, the role of the HBV infection process is currently actively developing the new HBV infections in the vitro model systems.
In our work the NTCP receptor for the expressing cell was used to obtain an efficient Semliki Forest virus (SFV) replicon expression system. The NTCP protein for easy detection its C-end was added to the green fluorescent protein (EGFP). Using genetic engineering techniques, there were developed two plasmid constructions pEF4 NTCP-EGFP and pSFV1 / NTCP p-EGFP containing NTCP-EGFP gene. To study the NTCP expression, localization and its functionality after the construction administration of the BHK-21 and the HepG2 cells, they were analyzed by immunofluorescence techniques. Demonstration of the effective NTCP-EGFP gene expression and the NTCP-EGFP protein localization in the cell plasma membrane and their ability to connect the HBV preS1 peptide. | en_US |
