Atvārsnīšu simetrija un asimetrija dažāda vecuma tabakas (Nicotiana tabaccum L.) lapās

Maģistra darba mērķis bija noskaidrot tabakas lapu atvārsnīšu dvīņu šūnas simetrijas izmaiņas un asimetrijas veidošanos atkarībā no lapas vecuma.
Par dažāda vecuma lapām izvēlējās jaunas lapas (1-2 cm ), pieaugušas un vecas (10 -15 cm ) lapas. No lapām tika pagatavoti preparāti lāzera skenējošajai konfokālajai mikroskopijai. Mikroskopēšanas laikā no dažāda vecuma tabakas lapām kopumā ieguva 300 atvārsnīšu attēlu sērijas. Atvārsnītēm un to kodoliem noteica to garumu, platumu, laukumu un perimetru. Tika noteiktas hloroplastu skaita atšķirības un kodolu perimetru atšķirības starp atvārsnīšu dvīņu šūnām dažāda vecuma tabakas lapās. Atvārsnītes iedalīja trijās vecuma grupās: jaunas, pieaugušas un vecas atvārsnītes.
Rezultāti uzrādīja, ka, lapām novecojot, starp atvārsnīšu dvīņu šūnām pieaug hloroplastu skaita un kodolu perimetru atšķirības, veidojot atvārsnītēs asimetriju. Jaunu lapu atvārsnītēs ir izteikta kodolu perimetru un hloroplastu skaita simetrija, bet vecu lapu atvārsnītēs tikpat izteikta kodolu perimetru un hloroplastu skaita asimetrija starp dvīņu šūnām.
No 300 atvārsnītēm, kuras iegūtas no jaunajām, pieaugušajām un vecajām lapām, dominēja jaunās un pieaugušās atvārsnītes aizņemot 65 % un 33%. The objective of the master work was to establish stomatal simetry and dissymety depending on the age of tobacco leaves.
For different ages were choosen young (1 -2 cm), adult and old leafs (15cm). Preparations were made for the Confocal laser scanning microscope from the leaves. From different age Tobacco leaves in general was gained 300 stomata image series. The stomata and guard cell nulear lenght, width, area and perimeter was measured. As well was established the residual between chloroplast count and diference between nuclear perimeter in leaves of different ages. stomata were divided in three age groups: young adult and old stomata.
The results showed that leaves geting older, between stomata guard cells increased the diference of chloroplast count and the diference between the nulear perimeter increasing the dissymetry of stomata. In the stomata of young leaves the perimeter of nuleus and number of chloroplast simetry is expressed, but in the stomata of old leaves the dissymetry of nuclear perimeter and chloroplast count is expressed.
From 300 stomata derived from new, adult and old leaves, the young and mature stomata dominated occupying 65% and 33% of all stomata.
