Rekombinanta hepatīta B vīrusa core antigēna imunoloģiski pētījumi; imunizētu dzīvnieku imunogenitātes novērtējums

Darba mērķis bija noteikt HBcAg- ietvertas 63 nukleotīdus garas ODN sekvences ietekmi uz organisma imūno atbildes veidošanos un imunogenitāti, salīdzināt iegūtos rezultātus ar kontroles proteīniem – HBcAg un HBcAg- (proteīns, kam izņemta dabiskā E.coli NS, kas rodas ekspresijas laikā).
No imunizētām BALB/c laboratorijas pelēm tika iegūti asins serumi un orgānu (liesa, limfmezgli) šūnu kultūras. Tika noteikta un salīdzināta proteīnu spēja inducēt antivielu sintēzi, šūnu kultūru proliferācijas indeksi 96 stundas pēc restimulācijas ar HBcAg- in vitro, CD4+ un CD8+ šūnu subpopulāciju pieaugums un savstarpējā skaita attiecība. Izmantojot komerciālus testus tika noteikta arī liesu šūnu kultūru supernatantā esošā IFN-γ koncentrācija un citokīna sintezējošo šūnu skaits.
ODN sekvences iekļaušana HBcAg daļiņā, salīdzinot ar HBcAg, HBcAg- un kontroles grupām, sekmēja augstus anti-HBc antivielu titrus, izteiktu CD8+ šūnu skaita pieaugumu un IFN-γ sintēzes palielināšanos imunizēto peļu imūnās atbildes laikā. Ar HBcAg- + ODN imunizēto organismu šūnu proliferācija bija zemāka, nekā izmantojot HBcAg, bet augstāka nekā, ja organismi imunizēti ar HBcAg-. The aim of this study was to determine the effect of a 63 nucleotide long ODN sequence packaged in HBcAg- particles on the immune response and immunogenicity of immunised organisms, also compare these results with control proteins – HbcAg and HBcAg- (a protein with no E.coli nucleic acid inside).
Blood serums and cell cultures (from spleens and lymph nodes) were acquired from immunised BALB/c laboratory mice. Anti-HBc antibody titres, proliferation of cell cultures after 96 hour re-stimulation with HBcAg- in vitro and increase in CD4+, CD8+ cell count and ratio was determined. Also the amount of IFN-γ and cells producing the cytokine in the spleen cell cultures was determined using commercial kits.
Packaging the ODN sequence in the HBcAg particles, compared to HBcAg, HBcAg- and control groups, facilitated high specific antibody titres, pronounced growth of CD8+ cells and synthesis of IFN-γ. Proliferation of the cells obtained from mice immunised with HBcAg- + ODN was lower than those immunised with HBcAg, but higher than those acquired from mice immunised with HBcAg-.
