Brīvi dzīvojošo amēbu konstatēšana ūdensapgādes sistēmās, izmantojot kultivēšanas metodi un to saistība ar baktērijam

Galvenais darba mērķis bija izolēt brīvi dzīvojošas amēbas no paraugiem, kas tika ņemti no ūdensapgādes sistēmas un izstrādāt kultivēšanas metodi uz piemērotas barotnes. Pec vairākiem dažādu barotņu un metožu izmēģinājumiem tika konstatēts, ka piemērotākā ir amēbu pirmskultivēšana uz E.coli baktērijam ar tālāko kultivāciju šķidrā barotnē, kur arī noteikta piemērotākā uzturvielu koncentrācija. Otrais mērķis bija izvērtēt sakarību starp amēbu klātbūtni un Legionella pneumophila baktērijām pētāmajos ūdens paraugos. Tika konstatēts, ka visos ūdens paraugos, no kuriem bija izolētas L. pneumophila baktērijas, bija konstatētas arī brīvi dzīvojošas amēbas. Tomēr atsevišķos gadījumos amēbas bija konstatētas arī tajos paraugos, kur nebija konstatētas L. pneumophila baktērijas. Trešais darba mēķis bija identificēt izdalītās brīvi dzīvojošas amēbas ar molekulārās bioloģijas metodēm. Tika noteikti piemērotākie šķidrās barotnes glabāšanas apstākļi amēbu kultivēšanai, kas nebija minēts zinātniskās publikācijās. The main aim of the work was to isolate the free-living amoebae from the samples, which were taken from the water supply system and to develop a method to cultivate them on a suitable medium. After testing of different media and methods, it was found that the most appropriate for amoeba is precultivation with E. coli bacteria with further cultivation in liquid medium, where are also the most appropriate concentration of nutrients was determined. Second aim of the work was to evaluate the correlation between the presence of free-living amoeba and Legionella pneumophila bacteria in investigated water samples. It was found that all samples without exception, where was found L. pneumophila bacteria were also found free-living amoebae. But amoebae were also found in those samples where not ascertained presence of L. pneumophila bacteria. Third aim of the work was to identify the isolated free-living amoebae with molecular biology methods. The suitable storage conditions for amoeba cultivation liquid media were found, which was not mentioned in publications.
