Listeria monocytogenes transpeptidāzes Sortāzes A inhibitoru atlase izmantojot uz fluorescences rezonanses enerģijas pārnesi (FRET) bāzētu skrīningu

Darba mērķis ir atrast jaunus inhibitorus, kas kavē Listeria monocytogenes transpeptidāzes sortāzes A veikto hidrolīzes reakciju, inhibējot sortāzes A darbību. Šī mērķa sasniegšanai tika izveidota DNS konstrukcija, kas kodē Listeria monocytogenes sortāzes A katalītisko domēnu. Tā kā vislabāk izpētīta ir Staphylococcus aureus sortāze A, papildus tika izveidota tās katalītisko domēnu kodējoša DNS konstrukcija. Ekspresija tika sekmīgi veikta E. coli ekspresijas sistēmā, abi proteīni bija šķīstoši. Sortāzes inhibitori tika atlasīti izmantojot augstas caurlaidības skrīningu, kas tika balstīts uz fluorescences rezonanses enerģijas pārnesi (FRET). 705 potenciālo inhibitoro savienojumu skrīninga rezultātā tika atlasīti 3 savienojumi. Divi no atlasītajiem savienojumiem specifiski inhibē abas sotrāzes, un viens savienojums inhibē L. monocytogenes sortāzi ievērojami spēcīgāk par S. aureus sortāzi. Papildus tika noteikta atlasīto kandidātvielu antibakteriālā iedarbība L. monocytogenes un S. aureus baktērijām, kā arī noteikts vielu citotoksiskums eikariotu šūnām, izmantojot NIH 3T3 peļu embriju fibroblastu šūnas. The aim of this study was to find novel inhibitors, in order to prevent the hydrolysis reaction performed by Listeria monocytogenes transpeptidase sortase A. For this purpose the DNA construct, encoding the catalytic domain of L. monocytogenes sortase A was created. The most studied is Staphylococcus aureus sortase A, for this reason, a DNA construct, encoding its catalytic domain was made. Both proteins were soluble after expression in E. coli expression system. In order to find the inhibitors, the high throughput screening based on the fluorescence resonance energy transfer (FRET) selection system was used. After the screening of 705 drug-like compounds, 3 inhibitors were found. Two selected compounds were found to inhibit both sortases and one compound showed higher inhibition of L. monocytogenes sortase. Additionally, the antibacterial activities of the selected compounds were tested on L. monocytogenes and S. aureus bacteria, as well as the cytotoxicity on the eukaryotic cell line, NIH 3T3 mouse embryo fibroblasts.
