Dažādu sāļu ietekme uz piekrastes augu Triglochin maritima un Tragopogon heterospermus sēklu dīgšanu

Darba mērķis ir noskaidrot dažādu sāļu šķīdumu ietekmi uz jūrmalas āžloka un pūkainā plostbārža sēklu dīgtspēju. Veica trīs eksperimentus – pirmajā divas nedēļas diedzēja 2015. gadā ievāktas un stratificētas jūrmalas āžloka sēklas Petri traukos dažādu sāļu (NaCl, Na2SO4, KCl, K2SO4, MgCl2, MgSO4) pieaugošās šķīdumu koncentrācijās (25, 50, 100, 200, 400 mM), pēc diedzēšanas perioda neuzdīgušās sēklas skaloja destilētā ūdenī un uz divām nedēļām ievietoja Petri traukos ar destilētu ūdeni un novēroja dīgšanas atjaunošanos. Otrajā eksperimentā dažādās sāļu koncentrācijās diedzēja 2016. gadā ievāktas un stratificētas jūrmalas āžloka un pūkainā plostbārža sēklas. Jūrmalas āžloka sēklas diedzēja tādos pašos sāļos un koncentrācijās kā pirmajā eksperimentā, savukārt ierobežota sēklu daudzuma dēļ pūkainajam plostbārdim analizēja tikai nātrija un magnija sāļu ietekmi. Lai noskaidrotu osmotiskā stresa ietekmi uz dīgtspēju, gan jūrmalas āžloka, gan pūkainā plostbārža sēklas līdzīgi kā sāļu variantā diedzēja izoosmotiskas koncentrācijas polietilēnglikola šķīdumos, pēc diedzēšanas perioda neuzdīgušās sēklas skaloja ar ūdeni un novēroja dīgšanas atjaunošanos. Trešajā eksperimentā četras nedēļas diedzēja nestratificētas jūrmalas āžloka sēklas dažādos (NaCl, Na2SO4, KCl, K2SO4, MgCl2, MgSO4) sāļu šķīdumos50 mM koncentrācijā, pēc diedzēšanas perioda neuzdīgušās sēklas skaloja ar ūdeni, diedzēja trīs nedēļas un novēroja dīgšanas atjaunošanos attiecībā pret kontroli. Pirmajos divos eksperimentos novēroja sakarību, ka, paaugstinoties sāļu koncentrācijai, samazinās sēklu dīgtspēja. Jūrmalas āžloka sālsizturība ir lielāka nekā pūkainajam plostbārdim, taču abas augu sugas ir izturīgas pret osmotisko stresu. Pirmajā eksperimentā apstrāde ar zemas koncentrācijas sāļu šķīdumiem veicināja dīgšanas stimulāciju pēc skalošanas ar ūdeni. Trešajā eksperimentā novēroja tendenci uz nestratificētu sēklu dīgšanas stimulāciju sāļu apstrādes rezultātā, taču nepieciešami turpmāki pētījumi, lai to apstiprinātu. The aim of this study is to find out the effect of sodium, potassium and magnesium salts on germination of seeds of the seaside arrowgrass and Tragopogon heterospermus. Three experiments were conducted. The first experiment was performed with seeds of seaside arrowgrass collected in the 2015 and stratified before germination. Seeds were germinated in Petri dishes for two weeks in solutions of different salts (NaCl, Na2SO4, KCl, K2SO4, MgCl2, MgSO4) in increasing concentrations (25, 50, 100, 200, 400 mM). After the first germination period ungerminated seeds were transferred to distilled water for two more weeks to observe germination recovery. The second experiment was conducted with seaside arrowgrass and T. heterospermus seeds that were collected in the 2016 and stratified before germination. The seeds of the seaside arrowgrass were germinated in the same solutions and concentrations as in the first experiment, however due to limited resources T. heterospermus seeds were germinated only in sodium and magnesium solutions. To assess the effect of osmotic stress, seeds of both species were germinated in iso-osmotic concentrations of polyethyleneglycol. After the germination period ungerminated seeds were transferred to distilled water for two more weeks to observe germination recovery. The third experiment was conducted with unstratified seeds of the sea arrowgrass in different (NaCl, Na2SO4, KCl, K2SO4, MgCl2, MgSO4) salt solutions (50 mM). The seeds were germinated for four weeks and after the germination period ungerminated seeds were transferred to distilled water for three more weeks to observe germination recovery in relation to control. The conclusion of the first two experiments was that increasing salinity decreases germination and that the sea arrowgrass has higher salt tolerance than T. heterospermus; however. both species are equally resistant to osmotic stress. Stimulation of germination after exposure to low concentrations of salt solutions was observed in the first experiment. A tendency of germination stimulation was observed in the third experiment, but the data was inconclusive. Key words: seaside arrowgrass, Tragopogon heterospermus, seed germination, salt tolerance.
