Trīs jaunu Siphoviridae dzimtai piederošu bakteriofāgu genoma un proteoma raksturošana

Abstract
Darbā pētīti trīs Siphoviridae bakteriofāgi no BMC DNS-fāgu kolekcijas. Papildus molekulārās bioloģijas metodēm darbā plaši lietotas in silico metodes. Iegūtās fāgu genomu consensus sekvences tika validētas ar atbilstošiem eksperimentāliem datiem. Fāgu genomi tika anotēti un tie uzrādīja augstu oriģinalitātes pakāpi. Tika konstatēts, ka jauniegūto fāgu proteomos atrodami daudzi evolucionāri konservatīvi ORF produkti, kuru homologi sastopami dažādos organismos, bet kuru funkcija paliek nezināma, līdz ar to tie kalpo par kandidātiem turpmākiem proteīnu strukturāli-funkcionāliem pētījumiem. in silico modulēšana ļāva izvēlēties turpmākās in vitro darbības genomu fizisko galu un DNS pakošanas stratēģijas noteikšanai. Pētīto fāgu genomu sekvences šobrīd tiek gatavotas deponēšanai gēnu bankā.
 
Three newly-isolated Siphoviridae bacteriophages from BMC DNA bacteriophage collection were studied. In addition to traditional molecular biology techniques, in silico methods were extensively used during this work. Consensus sequences of phage genomes were validated with subsequent experimental data. Genomes were annotated and showed high degree of novelty as of today. It was found that phage proteomes contain many evolutionary conserved ORF products with the homologues in different organisms, however, their functions yet remain unknown and therefore they might be considered noteworthy candidates for future functional-structural studies of proteins. In silico methods have hinted at the actions needed for determination of phage genome termini in vitro. Genomes of studied phages are currently getting ready to be deposited in gene bank.
