dc.description.abstractŠī darba ietvaros tika pētīta HHV-6 un CCL5 iekaisuma hemokīnu iesaistes loma autoimūnā tireoidīta attīstībā. Pētījumā iesaistīti 60 pacienti ar AIT, kuriem veikta tireoīdektomijas (Thyreoidectomia totalis) operācija, 30 autopsiju paraugi un 30 veseli asins donori kā kontroles grupa. HHV-6 genoma sekvence tika atrasta 59 (98%) AIT pacientiem gan audu, gan asins DNS paraugos un 23 (77%) kontroles grupas pacientiem. Aktīva infekcija tika noteikta 35/59 (59%) AIT pacientu vairogdziedzeru audu paraugos. Vīrusa slodze pie aktīvas infekcijas bija lielāka kā pacientiem pie latentas infekcijas (mediāna 1206 IQR: 455-2804 pie aktīvas pret 544.5 IQR: 155-1589 kopijas/1x106 šūnām pie latentas infekcijas) AIT pacientiem RANTES līmenis bija statistiski zemāks (mediāna 9.267, IQR: 3.489-16.93) kā asins donoriem (mediāna 56.6, IQR: 30.71 - 101.5) AIT pacientiem ar aktīvu infekciju, kuriem tika detektēta HHV-6 U12 un/vai U51 gēnu ekspresija, uzrādīja daudz zemāku RANTES līmeni, nekā pacientiem ar aktīvu vīrusa infekciju bez U12 un U51 gēnu ekspresijas. AIT pacientu audiem un asinīm, kā arī kontroles grupas asins paraugiem veica imunohistoķīmiju. Kontroles grupas asins donoru paraugiem un AIT pacientu audu paraugos tika konstatēta RANTES un HHV-6 gB (gp116) ekspresija. AIT pacientu PBMC paraugos tika konstatēta un HHV-6 gB (gp116) ekspresija, turpretim RANTES ekspresija konstatēta ļoti vāja.
dc.description.abstractThe aim of this study was to evaluate the potential role of human herpesvirus 6 (HHV-6) and CCL5 involvement in AIT development. This study included 60 AIT patients, 30 autopsied subjects without thyroid pathologies and 30 healthy donors as controls. HHV-6 genome sequence was detected in 59 (98%) AIT patients’ thyroid gland tissue and blood DNA samples and 23 (77%) in control group. Active infection was detected in 35/59 (59%) AIT patients’ thyroid gland tissue. Higher median viral load was found in patients with active viral infection than in patients with latent infection (median 1206 IQR: 455-2804 active and 544.5 IQR: 155-1589 copies/1x106 cells latent infection). AIT patients’ RANTES level was statistically lower (median 9.267, IQR: 3.489-16.93) compared to blood donors (median 56.6, IQR: 30.71 - 101.5) AIT patients who were positive for HHV-6 U12 and/or U51 gene expression, showed much lower RANTES level, compared to patients with active viral infection without the expression of U12 and U51 genes. RANTES and HHV-6 (gp116) was visualised in AIT patients’ tissue and blood, as well as control groups blood samples with immunohistochemistry. RANTES and was detected in control groups’ blood donor samples and AIT patient samples. HHV-6 gB (gp116) expression was detected in AIT patients’ PBMC samples, however RANTES expression was relatively low.
