Transkriptoma analīze akromegālijas pacientiem ar un bez medikamentozās terapijas

Darba mērķis bija veikt somatotrofas hipofīzes adenomas audu transkriptoma analīzi pacientiem, kuri ir vai nav lietojuši medikamentozo terapiju pirms operācijas, analizējot SST/DA analogu izraisītās izmaiņas transkriptomā. Lai to realizētu tika izdalīts totālais RNS no pacientu somatotrofās hipofīzes adenomas audiem un sagatavotas transkriptoma bibliotēkas. Pēc tam iegūtie dati tika analizēti, izmantojot Galaxy platformu un programmu R un R Studio. Pēc datu analīzes tika iegūti 22 statistiski būtiski DEGs un no tiem viens būtiski iesaistīts signālceļš, kā arī intensitātes grafikā tika novērotas divas izteiktas gēnu grupas, kas iedalijās starp pacientiem atkarībā no medikamentozās terapijas. The purpose of the study was transcriptome analysis of somatotroph pituitary adenoma tissue samples from the patients with and without medical therapy before surgery, by analysing SST/DA analogue caused changes in the transcriptome. To do this, total RNA was extracted from the tissues of patients’ somatotrophic pituitary adenoma and transcriptome libraries were prepared. The obtained data were analyzed using the Galaxy platform and the R and R Studio program. Following the data analysis, 22 statistically significant DEGs and from them one significantly involved signal pathway were identified, and two distinct groups of genes were observed in the heatmap, divided between patients depending on the treatment.
