Darbs veltīts vairāku endogēno vielu – γ-melanokortīnu peptīdu (γ-MSH), aminoskābes taurīna, un eksogēno dabas vielu – peptīda lunasīna, kartupeļu sulas un triterpēnu (betulīna, betulīnskābes un lupeola) ietekmes uz CNS pētījumiem, nosakot in vivo to darbības mehānismus un ietekmi uz peļu uzvedības reakcijām. Pirmo reizi parādījām, ka peptīdiem γ1- un γ2-MSH piemīt anksiogēna darbība. Lunasīns izraisa centrālas darbības hipolokomotorus efektus, kas daļēji realizējas caur dopamīna D1 receptoru. Aminoskābe taurīns mazās devās GABA-A receptoru līmenī ir aktīvāka par sintezēto taurīnu saturošo dihidropiridīnu atvasinājumu tauropironu. Augu valsts produktiem kartupeļu sulai un betulīnam piemīt antikonvulsanta un GABAerģiska aktivitāte. Atrastie dabas un endogēno vielu darbības mehānismi norāda uz iespējām jaunu psihotropo zāļu vielu dizainam. The aim of the Thesis was to assess the influence on the CNS functions of endogenous substances - γ-melanocortins (γ-MSH peptides), amino acid taurine, and the natural exogenous products - peptide lunasin, potato juice and triterpenes (betulin, betulinic acid, lupeol). The influence on behavior in mice and mechanisms of action were studied.
For the first time, anxiogenic action of γ1- and γ2-MSH and lunasin-induced central hypolocomotion provided at least in part via dopamine D1 receptor, were identified. Taurine exerted higher activity at the level of the GABA-A receptor than taurine-containing dihydropyridine derivative - tauropyrone. Potato juice and betulin possessed GABAergic activity and anticonvulsant action. Demonstrated effects of natural products and endogenous substances can be useful for the novel psychotropic drug design.
