Purīna patēriņš maizes rauga Saccharomyces cerevisiae purīna sintēzes mutantos

Maizes raugs ir lielisks eikariotu metabolisma pētījuma objekts, kas var stipri uzlabot izpratni par purīna metabolismu, un tā iespējamiem praktiskiem pielietojumiem. Šī darba ietvaros purīna biosintēzes de novo metaboliskā ceļa mutanti tika audzēti purīna limitācijas apstākļos 96 lauciņu platē. Iegūtās augšanas līknes tika analizētas, lai iegūtu informāciju par periodisko kultūru kinētiskiem parametriem, substrātu afinitāti un purīnu iekššūnas rezervēm. Tika noskaidrots, ka adenīna auksotrofiem ir lielāka afinitāte pret āršūnas adenīnu, salīdzinot ar hipoksantīnu. Āršūnu hipoksantīns ir pārsvarā izmantots purīnu rezervju veidošanā, kas bija lielākas CEN.PK celmā. Purīna biosintēzes ceļa sākuma posma mutanti patērēja purīnu efektīvāk, kas varētu norādīt uz to lomu metabolisma ceļa regulācijā. Baker’s yeast is a great tool for studies of eukaryotic metabolism and can greatly improve understanding of purine metabolism and its possible practical applications. In this work mutants of the purine biosynthesis de novo pathway were grown in purine limiting conditions in a 96-well plate. The obtained growth curves were analysed to obtain information about batch culture kinetic parameters, substrate affinity and purine intracellular reserves. It was found that adenine auxotrophs have a higher affinity for extracellular adenine, compared to hypoxanthine. Extracellular hypoxanthine is mainly used for creation of purine reserves, which are also higher in CEN.PK strain. Mutants of the first steps of the purine biosynthesis pathway were found to have a more effective purine consumption that could indicate their role in pathway regulation.
