Sadalīšanās produktu noteikšana Bikarfen 50mg tabletēs, izmantojot AEŠH metodi

Literatūras apskatā apkopota informācija par Bikarfen 50 mg tablešu sastāvu, izmantošanu farmācijā un to aktīvi farmaceitisko vielu - sehifenadīna hidrohlorīdu. Aprakstīti degradācijas pētījumi, to pieejamās vadlīnijas un veidi – hidrolīze, oksidēšana, termiska apstrāde un fotolīze. Darbā ir apkopota informācija par augsti efektīvās šķidrumu hromatogrāfijas (AEŠH) metodes validāciju. Darba eksperimentālajā daļā tika veikta Bikarfen 50 mg tablešu, Bikarfen 50 mg tablešu placebo un sehifenadīna hidrohlorīda degradācija ar skābi, bāzi un ūdeņraža peroksīdu. Analizējamo paraugu degradācijas pētījumi tika veikti arī “saules” gaismā, mitrumā un paaugstinātā temperatūrā. Iegūtie degradācijas produkti tika atdalīti no sehifenadīna hidrohlorīda, izmantojot AEŠH metodi. Joslu tīrības novērtēšanai tika izmantots diožu matricas detektors; masas līdzsvars tika aprēķināts, lai pārliecinātos, vai visi sadalīšanās produkti atdalās no sehifenadīna hidrohlorīda pamatjoslas un to saturs ir vienāds ar sehifenadīna hidrohlorīda satura samazinājuma daudzumu. Nezināmie piemaisījumi tika identificēti, izmantojot AEŠH-AIMS. AEŠH metodes validācija tika veikta pēc Eiropas Farmakopejas un ICH vadlīnijām. Literature review summarizes the information on the composition of Bicarfen 50 mg tablets, their use in pharmacy and their active pharmaceutical ingredient – sequifenadine hydrochloride. Also, HPLC methods validation and degradation studies are described, their available guidelines and types – hydrolysis, oxidation, thermal and photolytic degradation. Degradation of Bicarfen 50 mg tablets, Bicarfen 50 mg tablets placebo and sequifenadine hydrochloride with acid, base and hydrogen peroxide were performed. Forced degradation studies of analyzed samples were also performed using relevant conditions (light, heat and humidity). Degradation products were seperated from sequifenadine hydrochloride using high performance liquid chromatography. The estimation of peak purity was determined using diode array detection; mass balance calculations were made to determine if all degradants are accounted. Unknown impurities were identified using HPLC-HRMS. The validation was conducted in accordance with the European Pharmacopoeia and International Conference of Harmonization guidelines.
