Vides DNS molekulārā noteikšana ir metode, ar kuras palīdzību ir iespējams identificēt organismus pēc to ģenētiskā materiāla kopuma, ko ievāc no vides parauga. Ar šo metodi varētu papildināt tradicionālo zivju monitoringu vai aizvietot to pilnībā vērtējot reto zivju sugu sastopamību. Darba mērķis ir izstrādāt uz vides DNS noteikšanu balstītu metodiku retu un invazīvu sugu identificēšanai Latvijas ūdenstilpēs. Darbā tika ievākti 36 sugu zivju, vēžu un nēģu audu paraugi, lai izveidotu mtDNS COI gēna sekvenču datu bāzi. Darba ietvaros tika veikta vides DNS detekcija ūdens paraugos ar rlPĶR, kam tika izstrādāti un pārbaudīti praimeri un zondes deviņām reti sastopamām vai invazīvām zivju un vēžu sugām Latvijā, pamatojoties uz citohroma oksidāzes 1 gēnu. Metodikas testēšanā dabā izdevās identificēt retās un invazīvās sugas. Environmental DNA molecular detection is a method by which organisms can be identified from the totality of their genetic material collected from an environmental sample. This method could complement traditional fish monitoring or replace it by fully assessing the occurrence of rare fish species. The aim of the work is to develop a methodology based on the determination of environmental DNA for the identification of rare and invasive species in Latvian water bodies. Tissue samples from 36 species of fish, crayfish and lamprey were collected to create a database of mtDNA COI gene sequences. Within the framework of the work, environmental DNA detection was performed in water samples with rtPCR, for which primers and probes were developed and tested for nine rare or invasive fish and crayfish species in Latvia based on the cytochrome oxidase 1 gene (mtDNA-CO1). Rare and invasive species have been identified in field testing.
