VLP-based vaccines elicit a strong antibody response in the body, enhanced by antigen motifs arranged and repeated symmetrically on the surface of VLPs. The aim of the bachelor's thesis was to develop prototypes of VLP-based anti-sclerostin vaccines that could potentially inhibit the endogenous sclerostin self-antigen in the body. In the bachelor’s thesis sclerostin expression vectors were constructed, the recombinant sclerostin protein was expressed in E. coli, purified and chemically conjugated to different VLPs by two different methods. The most effective conjugation with native sclerostin was found to be in Qβ and ESE001 VLPs using the chemical linkers BCN-PEG8-NHS and tetrazine-NHS, so these conjugates were selected as potential anti-sclerostin vaccine prototypes.
