Šajā darbā tiek apskatīts kadherīns-11 un VE-kadherīns, un kāpēc šo proteīnu pētīšana ir racionāls virziens terapeitisko zāļu izstrādē. Darbā tika izveidoti konstrukti VE-kadherīnam un kadherīnam-11, divi saturēja veselus EC1-EC2 domēnus (VEFL, 11FL) un divi ar EC1 domēna delēciju (VE*, 11*). Pirmais darba mērķis, attīrīšanas metodes optimizācija, tika paveikts ar optimālu beigu iznākumu konstruktiem - 11*, 11FL, VE*. Nākošais mērķis, kadherīna-11 saistītspējīgu fragmentu atlase, tika veikts ar DSF metodi, iegūstot 75 potenciālus fragmentus. Pēdējais mērķis, fragmentu validācija, veikts ar NMR waterLOGSY metodi, atlasīja trīs saistītspējīgus fragmentus, bet nākošie testi, ITC un kristālu mērcēšana, neapstiprināja nevienu no 75 fragmentiem kā kadherīna-11 saistītspējīgu. This thesis focuses on cadherin-11 and VE-cadherin, and why targeting these proteins might be a useful approach to develop pharmaceutical therapies. For the targeting of VE-cadherin and cadherin-11, two constructs were made containing full EC1-EC2 domains (VEFL, 11FL) and two with truncated EC1 domain (VE*, 11*). The first aim was to optimise the purification procedure which was accomplished with optimal yields of constructs - 11*, 11FL, VE*. Another objective, the identification of fragment binders was screened using the DSF method and yielded 75 fragment hits of 11*. The last aim, validation of potential hits by an NMR waterLOGSY test revealed three potential binders which was continued using ITC and crystal soaking, indicating that none of the 75 fragment hits were binders of 11*.
