Abstract
Crypthecodinium cohnii ir mikroaļģe, kas producē omega-3 taukskābi DHA. Šajā darbā izveidots C. cohnii metabolisma kinētiskais modelis, kurā iekļauta centrālā metabolisma daļa acetil koenzīma-A (AcCoA) producēšanai citosolā. AcCoA C. cohnii ir DHA prekursors. Modelis spēj simulēt AcCoA producēšanu C. cohnii, izmantojot glikozi, glicerīnu vai etanolu. Modelis tika izveidots un analizēts programmā COPASI. Modeļa parametru iegūšanai tika izmantoti glikozes, glicerīna un etanola uzņemšanas ātrumi, kā arī 13C glikozes fluksomikas dati. Ar COPASI Sensitivities funkciju tika noteikti AcCoA produkciju limitējošie enzīmi. Izmantojot glicerīnu un glikozi, limitējošie enzīmi bija līdzīgi, taču, izmantojot etanolu, tie atšķīrās no citiem substrātiem.
 
Crypthecodinium cohnii is a microalgae that produces the omega-3 fatty acid DHA. In this study, a kinetic model of metabolism for C. cohnii was created, which incorporates the central metabolism needed for acetyl coenzyme-A (AcCoA) production in the cytosol. AcCoA is a precursor of DHA in C. cohnii. The model can simulate AcCoA production in C. cohnii using glucose, glycerol, and ethanol. The model was created and analyzed in the COPASI software. Glucose, glycerol, and ethanol uptake rates, as well as 13C glucose fluxomics data were used to obtain the model parameters. The COPASI Sensitivities task was used to determine the rate limiting enzymes for AcCoA production. The rate limiting enzymes were similar when using glycerol and glycose, however, they differed in the case of ethanol.
