Ar MYBPC1 saistīta miogēnā tremora peļu modeļsistēmas funkcionālā un molekulārā raksturošana

Lēnā tipa miozīnu saistošais proteīns-C (sMyBP-C), ko kodē MYBPC1 gēns, nodrošina resno pavedienu stabilizēšanu, kā arī regulē aktīna-miozīna saistīšanos. Missense varianti sMyBP-C M-motīvā, tostarp p.Y247H, pēdējo pāris gadu laikā tika saistīti ar iepriekš neaprakstītu iedzimtu miopātijas fenotipu – miogēno tremoru. Maģistra darba mērķis ir veikt cilvēku patogēnajam variantam p.Y247H MYBPC1 gēnā atbilstošās peļu modeļsistēmas funkcionālo un molekulāro raksturošanu. Darba gaitā veiktā funkcionālā analīze atklāja, ka ar slimību saistītais vājums ir salīdzinoši viegls. Veiktie molekulārie pētījumi, kas ietvēra transkriptomu analīzi, digitālo pilienu PCR un imūnblotu, atklāja, ka miogēnā tremora slimības mehānisms ir vairāk saistīts tieši ar muskuļu kontrakcijas funkciju izmaiņām. The myosin binding protein-C slow (sMyBP-C), encoded by the MYBPC1 gene, provides stabilization of thick filaments, and regulates actin-myosin binding. In the last few years, missense variants in the sMyBP-C M-motif, including p.Y247H, have been associated with a previously undescribed inherited myopathy phenotype - myogenic tremor. The aim of the master's thesis is to perform functional and molecular characterization of the myogenic tremor mouse model, corresponding to the pathogenic variant p.Y247H in the MYBPC1 gene. During the study, functional analysis revealed that the weakness associated with the disease is relatively mild. Molecular investigations, including transcriptome analysis, digital droplet PCR, and immunoblot, revealed that the mechanism of myogenic tremor disease is related more to direct alterations in muscle contraction function.
