Antimikrobiālā aktivitāte un heterologu antimikrobiālu peptīdu ekspresijas iespējas Zymomonas mobilis.

Zymomonas mobilis ir atpazīstama ar tās augsto etanola produkciju. Darba ietvaros tiek raksturota baktērijas antimikrobiālā aktivitāte un tās bioķīmiskā izcelsme. Tika noskaidrots, ka antimikrobiālās aktivitāte ir saistīta ar aerobā metabolisma blakus produktiem – acetātu un propionātu. Pēc tam veidoti mutanti celmi, lai samazinātu šo skābju sintēzi un novērotu skābju profila izmaiņas. Z. mobilis tika apskatīts kā potenciāls heterologu antimikrobiālu peptīdu producents tā atjūgtā metabolisma dēļ. No sākuma tika veikts piemērotu promoteru izvērtējums ar GFP, kur II tipa NADH dehidrogenāzes (ndh) promoteris bija visspēcīgākais. Tālāk ZM6 celmā tika veikta jauna antimikrobiāla peptīda - R10 - heterologa ekspresija zem ndh promotera, konjugējot to ar GFP kā nesējproteīnu, histadīna afinātes tagu un TEV proteāzes šķelšanas vietu. Zymomonsa mobilis is known for its high ethanol production. Here the antimicrobial activity and its biochemical origin are characterized. It was found that the antimicrobial activity is related to the by-products of aerobic metabolism - acetate and propionate. Mutant strains were then constructed to reduce synthesis of these acids and to observe changes in the acid profile. Z. mobilis was considered as a potential producer of heterologous antimicrobial peptides due to its uncoupled metabolism. Firstly, a screening of suitable promoters was performed with GFP, where the type II NADH dehydrogenase (ndh) promoter was found to be the strongest. Next, a novel antimicrobial peptide - R10, was expressed heterologously under the ndh promoter in the ZM6 strain, conjugated to GFP as a carrier protein, a histadine affinity tag and a TEV protease cleavage site.
