Šis pētījums apskata iespēju, izmantojot orgānu-uz-čipa (OOC) tehnoloģiju, pētīt kompleksas, cilvēka anaerobas mikrobiotas un tās komunikācijas mehānismus, kā, piemēram, ekstracelulārās vezikulas (BEV), ko varētu nākotnē lietot lai noskaidrotu mikrobiotas komunikācijas lomu audzēja patoģenēzē. Specializētā zarnas-uz-čipa (GOC) modelī tika kultivēta anaeroba zarnu mikrobiota un analizēts tās metagenoms, kā arī izdalīto BEV mazo RNS profils, kas spēj no zarnu lūmena nokļūt endotēlija cirkulācijā. Rezultāti norāda uz GOC modeļa spēju atainot funkcionālu zarnu modeli, un sniegt fizioloģiski nozīmīgu ieskatu anaerobas mikrobiotas un to izdalītu BEV dinamikā. This study examines the possibility of using organ-on-a-chip (OOC) technology to investigate the complex mechanisms of human anaerobic microbiota and its communication, such as extracellular vesicles (BEV), which could be used in the future to understand the role of microbiota communication in tumor pathogenesis. In a specialized gut-on-a-chip (GOC) model, anaerobic gut microbiota was cultured and analysis performed on metagenome, as well as the profile of small RNAs released in BEVs. The results indicate the ability of the GOC model to represent a functional gut model and provide a physiologically significant insight into the dynamics of anaerobic microbiota and their released BEVs.
