Išēmiskā insulta ārstēšana ietver limitētas terapijas opcijas. Ja radušos simptomu novēršana nav izdevusies, insultu pārcietušajiem pacientiem attīstīsies dažādas smaguma pakāpes komplikācijas. Viena no biežākajām insulta komplikācijām ir motorie traucējumi, kas būtiski ietekmē pacientu dzīves kvalitāti. Eksperimentālie insulta modeļi, apvienojot tos ar dzīvnieku uzvedības testiem, paver iespēju jaunas, efektīvas insulta terapijas izpētei. Maģistra darba mērķis ir izvērtēt divu dažādu veidu dendrimēru nanodaļiņu kombināciju ar īsām interferējošām ribonukleīnskābēm (siRNS) efektivitāti samazināt pēc insulta motoro funkciju traucējumus laipas un atvērta lauka testā. Pētījuma pirmo dienu atvērtā lauka testa rezultāti parāda, ka pēc dendronizētu zelta nanodaļiņu ievades peļu lokomotorā aktivitāte bija līdzīga naive kontroles grupai. Laipas testa rezultāti parāda pētāmo vielu potenciālu mazināt insulta izraisītu ķermeņa pušu asimetriju. Nepieciešami tālāki pētījumi, kas sniegtu papildus pamatojumu šo vielu efektivitātei insulta radīto motoro seku mazināšanā. There are limited treatment options available for the treatment of ischemic stroke, and failure to prevent its symptoms contributes to the development of various complications in stroke patients. Motor deficits are among the most common stroke consequences, and they have an impact on patients' quality of life. Experimental stroke models, when combined with animal behavioral tests, open the door to researching new, effective stroke therapies. The aim of the master’s thesis is to evaluate the effectiveness of two different types of dendrimer nanoparticles in combination with small interfering ribonucleic acid (siRNS) in reducing post-stroke motor functions in the beam walk and open field tests. The results of the open field test on the first days of the study show that after the introduction of dendronized gold nanoparticles, the locomotor activity of the mice was similar to that of the naive control group. The results of the beam walk test show the potential of the investigated substances to reduce the asymmetry of the sides of the body caused by stroke. Further research is required, which would provide additional justification for the effectiveness of these substances in reducing the motor complications of stroke.
