Lentinula edodes ekstrakta ietekme uz Drosophila melanogaster kāpuru oksidatīvā stresa rezistenci

Darbā pētīta Lentinula edodes β-glikānus un polifenolus saturoša ekstrakta ietekme uz savvaļas līniju Normal, Canton-S, Origon-R un Berlin-K Drosophila melanogaster kāpuru oksidatīvā stresa rezistenci. D. melanogaster kāpurus audzēti standarta barotnēs ar L. edodes ekstrakta piedevu(2mg/ml). Papildus eksperimentālās grupas veidotas ar lentināna (0,4mg /ml) un gallskābes piedevām (0,015 mg/ ml un 0,3 mg/ml). Lai noteiktu stresa rezistenci, analizēti fizioloģiskie parametri - kāpuru mutes kustību intensitāte, izdzīvotība un imago ķermeņa izmērs, un bioķīmiskie rādītāji – katalāzes un superoksīda dismutāzes aktivitāte, reducētā glutationa daudzums un brīvo skābekļa radikāļu līmenis. Konstatēts, ka visas analizētās piedevas paaugstina oksidatīvā stresa rezistenci, normalizējot visus apskatītos fizioloģiskos un bioķīmiskos rādītājus. Sēnes ekstraktam novērota visizteiktākā ietekme, līdzīgi rezultāti ir iegūti arī lentināna grupā un gallskābes lielākās koncentrācijas grupā. L. edodes ekstrakta efekts novērots visās darbā apskatītajās D. melanogaster līnijās. The effect of Lentinula edodes extract containing β-glucans and polyphenols on oxidative stress resistance of Drosophila melanogaster larvae of the wild-type lines Normal, Canton-S, Origon-R and Berlin-K was investigated. D. melanogaster larvae were reared in standard culture medium supplemented with L. edodes extract (2mg/ml). In addition, experimental groups were formed with the supplementation of lentinan (0.4 mg/ml) and gallic acid (0.015 mg/ml and 0.3 mg/ml). In order to determine stress resistance, physiological parameters were analyzed - the intensity of larval mouth movements, survival and imago body size, and biochemical indicators - catalase and superoxide dismutase activity, the amount of reduced glutathione and the level of free oxygen radicals. After normalizing all physiological and biochemical indicators, all of the analyzed supplements increase oxidative stress resistance,. The most pronounced effect was observed for the mushroom extract, similar results were also obtained in the lentinan group and in the group with the highest concentration of gallic acid. The effect of L. edodes extract was observed in all D. melanogaster lines examined in the work.
