Proantocianidīniem bagātu augu ekstraktu pretiekaisuma īpašību novērtējums plaušu epitēlija šūnu un imūnšūnu kultūrās in vitro

Proantocianidīnu antioksidatīvās un imunomodulārās īpašības padara tos par potenciāli vērtīgiem dabiskiem līdzekļiem ar zemu toksicitāti, ko iespējams iegūt no Latvijā izplatītu un komerciāli nozīmīgu ogu krūmu atzarošanas un ražas ievākšanas blakusproduktiem, veicinot ilgtspējīgas ražošanas principu ieviešanu praksē. Kā viens no proantocianidīnu pielietojumiem tiek izskatīts to pielietojums ar elpceļu infekcijām saistīta iekaisuma mazināšanā. Lai izpētītu to potenciālu baktēriju un vīrusu radītu iekaisumu nomākšanai, nepieciešams veikt novērtējumu to imunomodulārajām īpašībām imūnšūnās un plaušu epitēlija šūnās, kā arī analizēt iespējamo ekstraktu avotu un ekstrakcijas metožu ietekmi uz bioloģisko aktivitāti. Rezultāti parāda proantocianidīnu spēju samazināt iekaisumu veicinošā citokīna (IL-6) sekrēciju, kā arī ietekmi uz pretiekaisuma interleikīna (IL-10) sekrēciju imūnšūnu kultūrā, kas inducēta ar imunostimulatoriem LPS un Poly I:C. Salīdzinot dažādas ekstrakcijas metodes, konstatētas spēcīgākas imunomodulatoras īpašības ekstraktiem, kas iegūti izmantojot etanolu kā šķīdinatāju, joprojām saglabājot zemu citotoksisku efektu uz šūnām. The antioxidant and immunomodulatory properties of proanthocyanidins make them potentially valuable natural agents with low toxicity, that can be sourced from pruning and harvesting the by-products of widely distributed and commercially significant berry bushes in Latvia, promoting the implementation of sustainable production principles. One of the applications of proanthocyanidins is their use in reducing inflammation associated with respiratory infections. To explore their potential in suppressing bacterial and viral-induced inflammation, it is necessary to evaluate their immunomodulatory properties in immune and lung epithelial cells and analyze the impact of extraction sources and methods on biological activity. Results have shown that proanthocyanidins can reduce the pro-inflammatory cytokine (IL-6) while increasing the concentrations of anti-inflammatory interleukin (IL-10) in immune cells induced by immunostimulants LPS and Poly I:C. Comparing different extraction methods, stronger immunomodulatory properties were found in extracts obtained using ethanol as a solvent, while still maintaining low cytotoxic effects on cells.
