Darba mērķis bija noteikt dihidropiridīna BayK 8644 atvasinājumu ietekmi uz cilvēka mezenhimālo šūnu kultūru, kas iegūta no skeleta muskuļa biopsijas. Iegūtā šūnu kultūra tika raksturota, novērtējot dažādu gēnu ekspresiju, salīdzinot ar šūnu kultūrām, kas iegūtas no cilveka ādas dermas slāņa un kaulu smadzeņu aspirāta.
Dihidropiridīnu ietekme tika pētīta, novērtējot dažādus aspektus: šūnu dzīvotspēju un proliferācijas, morfoloģijas, gēnu ekspresijas izmaiņas. Šūnu proliferācijas izmaiņas tika pētītas, izmantojot Latvijā unikālu metodi – xCELLigence sistēmu.
Darba rezultāti uzrāda no skeleta muskuļu biopsijas iegūto šūnu heterogenitāti, ekspresējot plašu gēnu profilu. Tiek novērota BayK8644 atvasinājumu V-1-87, V-6-28, V-6-87 spēja pastiprināt šūnu proliferāciju.

Cell culture was characterized evaluating expression of different genes by comparing with cell cultures obtained from human dermal layer and bone marrow aspirate.
Influence of dihydropyridines was studied evaluating different aspects: changes of viability and proliferation, morphology and gene expression of cells. Cell proliferation was studied by using an unprecedent method in Latvia – xCELLigence (Roche) system.
The results of the research shows heterogenity of cells obtained from skeletal muscle biopsy by expressing wide profile of genes. There is also observed increase of proliferation by BayK 8644 derivatives V-1-87, V-6-28, and V-6-87.
