Lunasīna satura mērīšana peļu smadzenēs ar šķidrumu hromatogrāfijas - masspektrometrijas tandēma metodi

Problēma:
Lunasīns ir peptīds ar pierādītu pretvēža un pretiekaisuma efektivitāti. Ir vairāki pētījumi par lunasīna pierādīšanu graudos, bioloģiskajos šķīdumos, bet pētījumu par lunsaīna noteikšanu smadzeņu audos nav. Iepriekš mēs ar ŠH/ESI-MS/MS metodi noteicām lunasīna saturu graudaugos. ŠH/ESI-MS/MS metodes pielietošana lunasīna noteikšanai smadzeņu audos nav pētīta.
Pētījuma mērķis:
Izmērīt lunasīna saturu peļu smadzenēs pēc tā intracisternālas ievadīšanas.
Metode:
Pelēm intracisternāli ievadīja sintētisko lunasīna peptīdu un veica uzvedības testus. Pēc tam peles dekapitēja, lai paņemtu smadzeņu paraugus, kuri tika analizēti ar ŠH/ESI-MS/MS.
Rezultāti:
Lunasīna saturs smadzenēs 1,5 h pēc i.c. ievadīšanas pelēm tika pierādīts divās augstākajās koncentrācijas. Pēc lunasīna ievadīšanas devā 1 mmols/peli tas ir 30,7 ± 0,92 nmol/g un pēc devas 10 mmoli/peli – 155,7 ± 14 nmol/g.

Lunasin is a peptide with demonstrated anti-cancer, anti-inflammatory properties. There are couple studies of lunasin quantification in grains, biological fluids, but there are not studies on detection of lunasin in brain tissues. Previously we have used LC/ESI-MS/MS for lunasin quantification in grains of cereals. The use of LC/ESI-MS/MS technique for lunasin detection in brain tissue has not been studied.
Aim of research:
To measure lunasin content in mouse brains after intracisternal injection.
Method:
The synthetic lunasin peptide was injected intracisternally in mouse, and after that the behavior tests were carried out. After that the brain tissue were taken and analyzed by LC/ESI-MS/MS.
Results:
The lunasin concentration after i.c. injection at the dose of 1 mmol/mouse was found to be 30,7 ± 0,92 nmole/g and after injection at dose of 10 mmol/mouse – 155,7 ± 14 nmole/g.
