dc.description.abstract | Ubikitīna – proteasomu sistēma ir multienzīmatisks proteāžu komplekss, kas regule dažādus šūnu procesus. Šis sistēmas disregulācija var būt iesaistīta tādos cilvēka patoloģiju rašanās mehanismos kā 2. tipa cukura diabēts, juvenīlais idiopātiskais artrīts un Greivsa slimība. Tas var būt atkarīgs no proteasomas gēnu strukturālām pārmaiņām, kuri kodē proteasomu apakšvienības. Šajā pētījumā mēs analizējam asociāciju starp PSMA6 gēna promotera poly(A) (c. –655 An) mikrosatelīta garuma izmaiņām un bronhiālo astmu (BA), 1. tipa cukura diabētu (T1DM) un aptaukošanos (APT) Latvijas populācijā. PSMA6 gēna poly(A) polimorfisms tika analizēts ar gadījuma / kontroles pētījumu un bioinformātiskām programmām.
Ir konstatēti genotipi, kas asociējas ar aptaukošanās un 1. tipa cukura diabēta paaugstinātu saslimšnas risku. Pētāmā lokusa A17 alēle ir riska alēle asociācijā ar bronhiālo astmu un aptaukošanos Latvijas populācijā. A20 alēles gadījumā ir paaugstināts saslimšanas risks ar 1. tipa cukura diabētu Latvijas populācijā. Svarīgi atzīmēt, ka mēs konstatējam gan riska, gan protektīvus genotipus un alēles pret šīm patoloģijām cilvēka genomā. | en_US |
dc.description.abstract | The ubiquitine proteasome multycatalytic protease complexes regulate a variety of cellular processes. Deregulation of this system implicated in several human pathologies, such as type 2 diabetes mellitus, juvenile idiopathic arthritis and Grave’s desease. It can depend on the structural variations in the genes encoding the proteasome subunits. In this study we have analyzed the association between bronchial asthma (BA), type 1 diabetes mellitus (T1DM) and adiposity (APT) in Latvians and variation of the polyA (c.– 655 An) microsatellite, locating in the promoter of PSMA6 gene. PSMA6 gene microsatellite polymorphism was analyzed with case/control study and via bioinformatical programms.
It was revealed that some genotypes are associated with the risk of adiposity and type 1 diabetes mellitus development. Allele A17 turned out to be associated with bronchial asthma and adiposity. In the same time allele A20 was associated with type 1 diabetes mellitus development. It should be noted that we observed protective genotypes and alleles for these pathologies in human genome. | en_US |
