Selenofēnkumarīnu un selenofēnhinolonu salīdzinošā ietekme uz HepG2 šūnu fermentatīvo antioksidatīvo sistēmu

Latvijas Organiskās sintēzes institūtā izstrādātas metodes jaunu selenofēn[3,2-c] un [2,3-c]kumarīnu un selenofēn[3,2-c] un [2,3-c]hinolonu sintēzei, kuriem pierādīts antiproliferatīvais efekts in vitro.
Darbā tika noteikts savienojumu inhibējošais efekts uz trim hepatomas HepG2 šūnu galvenajiem antioksidatīvajiem fermentiem (GPx, TrxR, SOD) ar mērķi noskaidrot savienojumu antiproliferatīvā efekta darbības mehānismus.
Pētījumā konstatēts, ka visi savienojumi ir efektīvi GPx inhibitori. TrxR aktivitāti inhibē selenofēnhinoloni un [3,2-c] aizvietotie selenofēnkumarīni, savukārt SOD aktivitāti ietekmē tikai divi no savienojumiem. Kopumā noteikts, ka selenofēnhinoloni ir aktīvāki antioksidatīvo fermentu inhibitori.
Viens no savienojumu antiproliferatīvā efekta darbības mehānismiem var būt saistīts ar spēju modulēt antioksidatīvo fermentu darbību. Latvian Institute of Organic Synthesis has developed a method for the synthesis of substituted selenopheno[3,2-c] and [2,3-c]coumarins and selenopheno[3,2-c] and [2,3-c]quinolones, which exhibit antiproliferative activity in vitro.
The inhibitory effect of these compounds on three main HepG2 cell antioxidant enzymes (GPx, TrxR, SOD) has been determined. The aim of the study was to determine possible mechanism of action of these compounds.
The results show that all compounds are effective GPx inhibitors. Selenophenoquinolones and [3,2-c] selenophenocoumarins inhibit TrxR, while only two compounds affect SOD. Overall, selenophenoquinolones showed a better inhibitory effect on the enzyme activity.
One of the antiproliferative activity mechanisms of these compunds could be associated with the ability to modulate the antioxidant enzyme activity.
