Bakteriofāga Enc34 replikācijas moduļa proteīnu ekspresija, attīrīšana un raksturošana

Abstract

Darbā tika veikta bakteriofāga Enc34 replikācijas moduļa proteīnu sekvenču analīze un hipotētisko proteīnu ORF3, ORF4, ORF5 un ORF6 eksperimentāla izpēte ar mērķi identificēt to funkcijas. Pētījumu rezultātā praimāzei ORF1 tika atklāts iepriekš neidentificēts helikāzes domēns un ORF8 tika re-anotēts kā resolvāze. Rekombinanti producēts ORF4 bija nešķīstošs, savukārt ORF3 un ORF5 – šķīstoši, un to sekmīga attīrīšana ļauj darbu turpināt ar proteīnu kristalizāciju. ORF6 un delēcijas variantam ORF6Δ21 tika noteiktas kristālstruktūras, kuru analīze kopā ar in vitro eksperimentiem parādīja, ka ORF6 ir ssDNS saistošais proteīns (SSB). Pētījumā pirmoreiz tika atklāta SSB C-terminālā reģiona nozīme ssDNS saistīšanas specifiskuma nodrošināšanai. Darbs tika izstrādāts Latvijas Biomedicīnas pētījumu un studiju centrā Dr. biol. A. This study reports sequence analysis of bacteriophage Enc34 replication module proteins and experimental work on hypothetical proteins ORF3, ORF4, ORF5 and ORF6 to identify their function. During the study, a previously undetected helicase domain was found in the primase ORF1, and ORF8 was reannotated as resolvase. Recombinant ORF4 was insoluble, while ORF3 and ORF5 were soluble and successfully purified, allowing to proceed with their crystallization. Crystal structures of ORF6 and a deletion variant ORF6Δ21 were determined, which together with in vitro assays showed that ORF6 is a ssDNA-binding protein (SSB). This is the first study that demonstrates the role of the SSB C-terminal region for ssDNA binding specificity.
