Doliholu identifikācija un to līmeņu salīdzināšana žurku nierēs pēc poliprenolu un atorvastatīna ievadīšanas

Latvijā poliprenoli tiek tirgoti kā uztura bagātinātāji, bet nav daudz zinātnisku pētījumu par to farmakoloģisko iedarbību. Iepriekšējie pētījumi in vivo ir apliecinājuši poliprenolu spēju samazināt statīnu izraisīto muskuļu vājumu laboratorijas dzīvniekiem. Darbojoties kā HMG-CoA reduktāzes inhibitori, statīni nobloķē mevalonskābes sintēzi un, iespējams, arī Q10, poliprenolu un doliholu sintēzi, un tiek pieļauts, ka muskuļu vājums ir tieši šo savienojumu līmeņu samazinājuma dēļ. Iespējams, ka eksogēni ievadīti poliprenoli veicina doliholu sintēzi. Darba mērķis ir ar šķidruma hromatogrāfijas masspektrometrijas (lidojuma laika (TOF)) metodi izpētīt, kāda tipa doliholi atrodas žurku nierēs, izmērīt to līmeņus un noskaidrot, vai poliprenolu un atorvastatīna ievadīšana ietekmē doliholu spektru un daudzumu. Iegūtie rezultāti liecina, ka doliholu līmenis indivīdiem ir ļoti atšķirīgs. Dominē D17, D18 un D19. Poliprenoli un arī atorvastatīns palielina doliholu spektru nierēs. Despite the lack of scientific research regarding the pharmacological effects of polyprenols, they are readily available on Latvian market as dietary supplements. Previous in vivo research has confirmed the ability of polyprenols to reduce statin induced muscle weakness in laboratory animals. Acting as HMG-CoA reductase inhibitors, statins inhibit the synthesis of mevalonate and perhaps also the synthesis of Q10, polyprenols and dolichols. Supposedly the decrease in these compound levels is the main cause for muscle weakness. It is possible that exogenously administered polyprenols contribute to the dolichol synthesis The purpose of this paper is to research what types of dolichols are present in rat kidneys, measure their levels and investigate if administration of polyprenols and atorvastatin affects the spectrum and amount of dolichols, using liquid chromatography mass spectrometry (time of flight) method. The results show that individuals have a wide range of dolichol levels, the most prominent ones are D17, D18 and D19. Polyprenols and atorvastatin increase the spectrum of dolichol in rat kidneys.
