Dažādu škīdinātāju mijiedarbība ar kolorimetrisko testu reaģentiem

Veicot dažādus eksperimentos var būt nepieciešams izmantot vielas ar dažādu šķīdību ūdenī un organiskos šķīdinātājos. Nereti precīzai eksperimenta norisei nepieciešams noteikts pH līmenis, ko nākas izmainīt oriģinālajā šķīdumā. In vitro eksperimentos vielu šķīdībai ir ļoti liela nozīme, jo ir virkne priekšnosacījumu eksperimenta veikšanai, kas izriet no pašu eksperimentu būtības. Piemēram, eksperimentos ar šūnu līnijām jāizmanto barotnes un piedevas ar pH 7,4 un tām jābūt šķīdumiem ūdenī, jo organiskie šķīdinātāji dažādās koncentrācijās nonāvē šūnas. Eksperimentos ar izolētiem enzīmiem vai audu membrānām arī vienmēr šķīdinātāju kontroles ir obligātas. Eksperimentos in vitro visbiežāk izmanto reakciju gaitā radušās krāsu (kolorimetriskā metode), luminiscences un fluorescences mērījumus. Darba mērķis: pētīt dažādu šķīdinātāju ietekmi uz krāsas reakciju acetilholīnesterāzes un brīvā radikāļa DPPH testos in vitro. Iegūtie rezultāti liecina, ka šķīdinātāju izvēle būtiski ietekmē kolorimetrisko testu rezultātus, kā arī to, ka askorbīnskābi var izmantot kā pozitīvās references vielu, salīdzinot citu vielu antiradikālo efektu. By concluding different experiments, it might be necessary to use substances with different solubility in water and organic solvents. For a precise experiment process you often need an exact pH level, which needs to be changed in the original solution. Substance solubility is very important for in vitro experiments, as there are several prerequisites for the conclusion of the experiment, which result from the essence of the experiments. For example, cell line experiments require nutrient solutions and additives with pH 7,4 and they have to be solutions in water, as organic solvents at different concentrations kill cells. Solvent control is also mandatory for experiments with isolated enzymes or tissue membranes. In vitro experiments most commonly use color (colorimetric method), luminescence and fluorescence measurements resulting from the reactions. Objective of the paper: to research the impact of different solvents on color reaction in acetylcholinesterase and free radical DPPH tests in vitro. The obtained results show, that solvent choice significantly impacts the colorimetric test results and how ascorbic acid can be used as positive reference substance, in comparison to other substance antiradical effect.
