Sigma 1 receptora ietekme uz peļu mezenhimālo stromas šūnu osteodiferenciāciju

Abstract
Mezenhimālajām stromas šūnām (MSŠ) piemīt osteodiferenciācijas potenciāls. Sigma-1 receptors (SigmaR1) ir transmembrānu proteīns, tā ietekme uz osteodiferenciāciju nav vēl zināma. Bakalaura darba mērķis bija noskaidrot SigmaR1 ietekmi uz peļu taukaudu MSŠ (T-MSŠ) osteodiferenciāciju. Osteodiferenciācijas norise un SigmaR1 ietekme uz osteodiferenciāciju tika analizēta, pētot osteoģenēzē iesaistīto gēnu ekspresiju peļu T-MSŠ ar qPCR metodi. Salīdzinot izslēgta SigmaR1 (S1R-/-) T-MSŠ pret WT (wild type) T-MSŠ gēnu ekspresiju pēc osteodiferenciācijas, transkripcijas faktora osterix ekspresija tika nozīmīgi palielināta, bet nediferencētā stāvoklī būtiskas izmaiņas netika novērotas.
 
Mesenchymal stromal cells (MSCs) possess an osteodifferentiation protential. Sigma-1 receptor (SigmaR1) is a transmembrane protein, its impact on osteodifferentiation is unknown. The aim of the bachelor's thesis was to acknowledge the impact of SigmaR1 on the mouse adipose mesenchymal stromal cells (ASCs) osteodifferentiation. To determine the ostedifferentiation process and SigmaR1 impact on osteodifferentiation, the expression of osteogenic genes in mouse ASCs were analized with qPCR method. Comparing knock out SigmaR1 (S1R-/-) ASCs against WT (wild type) ASCs gene expression after the osteodifferentiation, transcription factor osterix was significantly upregulated, but at non-differentiated state significant changes were not observed.
