Beta-amiloīda peptīdu īso analogu ietekmes uz beta-amiloīda plāgu sakopojumu kvantitatīvajiem radītājiem izvērtēšana in vivo Alcheimera slimības modelī

Alcheimera slimība (AD) ir neirodeģeneratīva slimība un biežākais demences cēlonis, kas raksturojas ar amiloīda-β (Aβ) plāgu izgulsnēšanos. Pēdējos gados veiktie pētījumi vērsti uz garo un vidējo Aβ peptīdu izpēti, taču nav datu par īso Aβ peptīdu efektiem in vivo AD modelī. Darba mērķis bija izmantojot AxioVision kvantitatīvi raksturot īso 1-6Aβ peptīdu A2T-h, A2T-m, WT-h, WT-m ietekmi uz Aβ plāgu sakopojumiem in vivo AD modelī. Kvantitatīvā analīze parāda, ka pētītie īsie peptīdi salīdzinot ar kontroles grupu neuzrādīja statistiski ticamas atšķirības, nosakot to ietekmi uz toksisko plāgu vidējo skaitu un izmēru izmaiņām. Ņemot vērā iegūtos rezultātus, var secināt, ka ir nepieciešama tālāka īso peptīdu efektivitātes izpēte, izmantojot arī citas devas un sekvences peptīdus. In recent years, several studies have focused on long and medium Aβ peptides research, however there are not any data about short peptides in in vivo mouse models of AD. The aim of the study was to quantitatively characterize the effect of 1-6Aβ short peptides A2T-h, A2T-m, WT-h, WT-m on plaque aggregations in in vivo model of AD using AxioVision. Quantitative analysis represents that short Aβ peptides did not show statistically significance data of their effect on the number and size of toxic Aβ plaques reduction. Based on the obtained results, it can be concluded that further research on efficacy of short peptides is needed, using other doses and sequences.
