Antidiabētiska līdzekļa metformīna neirorestoratīvo efektu izpēte eksperimentāli izveidotajā Alcheimera slimības modelī

levads. Glikozes metabolisma traucējumi un kognitīvie traucējumi ir vienas no agrīnām sporādiskās Alcheimera slimibas (sAD) pazīmēm. Agrīna terapeitiskā iejaukšanās varētu apstādināt kognitīvos traucējumus pirms sAD progresēšanas. Metformīns, visplašāk zināms pretdiabeta līdzkelis, ir uzrādījis spēju uzlabot kognitīvās funkcijas un glikozes metabolismu otra tipa diabēta pacientos un diabēta modeļdzīvniekos. Intranazālais ievadīšanas veids ļauj samazinaāt preparāta devu un mazina blakusparadibu izpausmes. Pie tam, metformīns šķērso hematoencefālisko barjeru un ir spējīgs izplatīties smadzenēs pēc vienreizējās ievadīšanas. Intranazālas metformīna ievadīšanas efekti uz sociālo uzvedību iepriekš nav apskatīti sAD dzīvnieku modelī. Šī pētījuma mērķis bija noteikt intranazāli ievadīta metformīna efektus uz veselo un sAD tipa modeļžurku sociālo uzvedību. Šī pētījuma uzdevumi bija: 1) apkopot aktuālo literatūru par Alcheimera slimību, neiroprotektīviem metformīna efektiem un sociālo uzvedību; 2) veikt 3 istabu testu veselām un sAD tipa modeļžurkām un 3) analizēt un interpretēt iegūtos uzvedības datus. Metodes: metformīnu vadīja intranazāli (1 vai 3 mg/kg) kontroles vai sAD tipa pieaugušo tēviņu žurkām 28 secīgas dienas un 37. eksperimenta dienā veica 3 istabu testu. Iegūtie rezultati parāda, ka intranazāli ievadīts metformīns uzlaboja sAD tipa žurku sociālo interesi un vēlmi socializēties ar iepriekš nezināmu dzivnieku. Secinot, šie dati izceļ nepieciešamību tālākai izpētei, kas būtu vērsta uz metformīna efektu uz sociālo uzvedību molekulāro mehānismu noskaidroāanu sAD tipa modeļžurkās. Background. Impairments in glucose metabolism and cognition are one of the early signs of sporadic Alzheimer's disease (sAD). Early therapeutic interventions could prevent cognitive decline before sAD progresses. Metformin, the most widely used antidiabetic drug, has shown the ability to boost cognitive function and glucose metabolism in type 2 diabetes patients and diabetic model animals. Intranasal administration route allows for the decreased dose of administered drug and its side effects. Moreover, metformin crosses the blood-brain barrier and can distribute in the brain after a single administration. Effects of intranasal administration of metformin have not been previously studied in sAD-type animal model. The aim of this study was to determine the effects of intranasally administered metformin on social behaviour in healthy and sAD-type model male rats. The objectives of the study were: 1) to review current literature on Alzheimer’s disease, studies on neuroprotective effects of metformin and social behaviour; 2) to perform 3-chamber test on healthy and sAD-type model rats and 3) to analyze and interpret the obtained behavioural data. Methods: intranasal metformin (1 or 3 mg/kg) was intranasally administered to control or sAD- type adult male model rats for 28 consecutive days, and on experimental day 37 the 3-chamber test was performed. Obtained results show that intranasal metformin improved social interest and preference for social novelty of rats with sAD-type pathology. In conclusion, these data highlight the need for further research to clarify the molecular mechanisms of metformin action on social behaviour of sAD-type model rats.
