Nealkoholiskā taukaino aknu slimība NATAS ir visizplatītākā hroniskā aknu slimība pasaulē, un tās izplatība pieaug paralēli diabēta un aptaukošanās pieaugumam. NATAS ir raksturīga ar tauku uzkrāšanos aknās, kas pēc tam izraisa iekaisumu, kam var sekot citu aknu slimību attīstība – nealkoholiskais steatohepatīts (NASH), ciroze un hepatocelulārā karcinoma. Tomēr ietekme uz lipīdu uzkrāšanos un iekaisuma citokīnu veidošanos taukaino aknu šūnās nav labi izpētīta. Bakalaura darba mērķis ir pētīt dažadu zāļu vielu iedarbību uz pro-iekaisuma citokīnu IL6 un IL-8 veidošanos NATAS modelī, izmantojot HepG2 šūnas. Pētījuma rezultāti liecina, ka šim pētījumam izvēlētās aktīvās vielas ietekmē HepG2 šūnu dzīvotspēju atkarība no koncentrācijas un inhibē taukskābju izraisīto IL-6 un IL-8 sekrēcijas palielināšanos HepG2 šūnās pēc 24 h inkubācijas. Non-Alcoholic Fatty Liver Disease is the most common chronic liver disease in the world and its prevalence is increasing in parallel with the rise of diabetes and obesity. NATAS is characterized by the accumulation of fat in the liver, which then causes inflammation, which can lead to the development of other liver diseases - non-alcoholic steatohepatitis (NASH), cirrhosis and hepatocellular carcinoma. However, the effects on lipid accumulation and inflammatory cytokine production in fatty liver cells have not been well studied. The aim of the bachelor's thesis is to study the effects of various medicinal substances on the production of pro-inflammatory cytokines IL-6 and IL-8 in the NATAS model using HepG2 cells. The results of the study show that the active substances selected for this study affect the viability of HepG2 cells in a concentration-dependent manner and inhibit the fatty acid-induced increase in IL-6 and IL-8 secretion in HepG2 cells after 24 h of incubation.
