Bieži lietotu nenarkotisko pretsāpju līdzekļu ietekme uz dziļās glikēšanas galaproduktu veidošanos

Dziļās glikēšanas galaprodukti (AGE) tiek producēti reducējošo cukuru vai reaktīvo αoksoaldehīdu ne-enzimātiskā reakcijā ar biomakromolekulām. Bakalaura darba mērķis ir izpētīt bieži lietotu nenarkotisko pretsāpju līdzekļu (nimesulīda, diklofenaka, ibuprofēna, paracetamola, celekoksiba) ietekmi uz AGE veidošanos in vitro proteīnu glikēšanas modeļos, kas tiek plaši izmantoti potenciālo AGE inhibitoru efektivitātes noteikšanai. Iegūtie rezultāti rāda, ka no testētajiem nenarkotiskajiem pretsāpju līdzekļiem nimesulīdam piemīt visizteiktākie anti-AGE efekti, par ko liecina nimesulīda efektivitāte abos glikēšanas modeļos (ar metilglioksālu un glikozi). Ibuprofēns un diklofenaks uzrādīja anti-AGE efektus tikai testā ar glikozi. Paracetamolam un celekoksibam nevienā no glikēšanas reakcijām netika konstatēta AGE veidošanās inhibīcija. Advanced glycation endproducts (AGEs) are formed by non-enzymatic reaction of free reducing sugars or reactive α-oxoaldehydes with biomacromolecules. The aim of bachelor’s thesis is to evaluate the effects of frequently used non-opioid analgesics (nimesulide, diclofenac, ibuprofen, paracetamol, celecoxib) on the formation of AGEs in in vitro protein glycation models, which are widely used to determine the efficacy of potential AGE inhibitors. The results show that nimesulide has the most pronounced anti-AGE effects compared to the tested non-opioid analgesics, which is evidenced by the significant effectiveness of nimesulide in both glycation models. Ibuprofen and diclofenac showed anti-AGE effects only in the glucose test. Paracetamol and celecoxib showed no inhibition of AGE formation in any of the glycation reactions.
