Polipeptīdu racemizācijas un degradācijas pētīšana ar KMR metodi

Darbā tika veikta trifluoretiķskābes šķīdumu ietekmes izpēte uz cietfāzes sintēzē iegūtajiem polipeptīdiem. Tika pagatavoti pētāmo vielu paraugu šķīdumi un uzņemti to spektri ik pēc noteiktiem laika intervāliem, noteikti polipeptīdu sastāvā esošo aminoskābju αCH protonu ķīmisko nobīžu apgabali 1H KMR spektros. Tika izvērtēts, kāda ir trifluoretiķskābes šķīdumu ietekme uz oksitocīna, dalargīna un triptorelīna racemizāciju un stabilitāti. Tika noteikts, ka triptorelīns degradē 5,15% CF3COOH D2O šķīdumā, kā arī tiek pakļauts racemizācijai. Oksitocīns 5,15% CF3COOH D2O šķīdumā var veidot racēmisko maisījumu. Savukārt dalargīns ir stabils visos analīžu veikšanas apstākļos. Iegūto 1H KMR spektroskopijas datu atbilstības pārbaudei tika uzņemti AEŠH/MS spektri. This work entailed the studies of effect of trifluoroacetic acid solution on solid-state synthesized polypeptides. The solution samples were prepared and their spectra were obtained at specified time intervals and the polypeptide make up was ascertained from the αCH proton chemical shift region in 1H-NMR spectra. The effects of trifluoroacetic acid solution on the racemization and stability of oxytocin, dalargin and triptorelin .were noted. It was determined that triptorelin degrades in 5,15% CF3COOH D2O solution, and in addition, is suceptable to racemization. Oxytocin in 5,15% CF3COOH D2O solution formed a racemic mixture. Dalargin was stable under all experimental conditions used. HPLC/MS was used to confirm the 1H NMR results.
