dc.description.abstract | Kursa darbā apskatīta peptīda Triptorelīna cietfāzes sintēze, kurā par aktivēšanas reaģentu izmanto HCTU un HBTU un kā bāzi lieto diizopropiletilamīnu. Tuvāk aplūkota kondensācijas reakcija un peptīda saites veidošanās mehānisms. Par analītisko metodi izmantots ninhidrīna vai hloranila tests. Veidots acilēšanās reakcijas modelis datorprogrammā SPARTAN'04 Wavefunction Inc, ASV. Triptorelīna cietfāzes sintēzes produktā identificēti iespējamie piemaisījumi, kurus veido iztrūkstošas aminoskābes un dubultas aminoskābes peptīdā. Apskatīti iespējamie Triptorelīna diastereomēri, kuri veidojas, ja acilēšanas reacijā noris aminoskābes epimerizācija. Salīdzināti praktiski iegūtie kondensācijas reakcijas rezultāti ar teorētiski aprēķinātajiem. Produkts analizēts ar augsti efektīvo šķidrumu hromatogrāfiju, masspektroskpiju, ar 1H-KMR spektroskopiju, izmantojot divdimensionālos TOCSY spektrus. | en_US |
dc.description.abstract | Solid phase peptide synthesis of Triptorelin have been studied.The activation of the carboxylic acid function was carried out by using HCTU as a coupling reagent and di-isopropylethylamine as a base. The condensation reaction and peptide bond formation were looked at in more detail. The molecular modelling was carried out on SPARTAN'04 Wavefunction Inc, US with the purpose of predicting the relative facility of each amino acid addition.. Possible by-products of the Triptorelin solid phase synthesis were identified as insertation or deletion aminoacids in the peptide. The work also looks at the possible diastereomeres of Triptorelin that can form if an epimerization of an amino acid occurs during the acilation reaction. Experimentally acquired results of the condensation reaction were compared with theoretical calculations. The product was analysed with highly effective liquid chromatography, massspectroscopy, 1H-KMR spectroscopy, using 2D TOCSY spectrs. | en_US |
