Darba nosaukums ir „No klīniskā materiāla izdalīto stafilokoku īpašību izpēte.”

Šis darbs sevī ietver koagulāzes negatīvo stafilokoku (KONS), to fenotipisko un
ăenētisko īpašību, kas nosaka mikroorganismu virulenci, izpēti. Finansiālo atbalstu nodrošina
Latvijas ZinātĦu padomes pētījumu grants. Galvenā pētījumu daĜa tika veikta LU
Bioanalītisko metožu laboratorijā (vadītājs Dr. biol. Jānis Ancāns). Aparatūras iegāde
finansēta no Eiropas Savienības pētījuma granta ERAF.
Galvenie darba mērėi ir izdalīt gēnus un fermentus, kas nosaka virulenci, salīdzināt
to klātbūtni klīnisko paraugu grupā un kontroles grupā, kā arī noteikt saistību starp šiem
gēniem un mikroorganismu patogēniskumu.
Darbā izvirzītās problēmas būtība slēpjas KONS, tai skaitā tā galvenā pārstāvja
S.epidermidis, patogēniskuma potenciāla noteikšanā. S.epidermidis ir daĜa no normālas
cilvēka ādas mikrofloras, tomēr dažos gadījumos tas parādās kā nozokomiālais patogēns.
Parasti S.epidermidis izraisītās infekcijas attīstās uz pazeminātas imunitātes fona, kā arī ir
saistītas ar medicīnisko iekārtu izmantošanu. Šajā darbā ietvertais pētījums tāpat parāda
virulences gēnu ica un aap lomu biofilmu veidošanās mehānismā un nozokomiālo stafilokoku
izplatībā.
Darbā tika izmantota paraugu identifikācijas bakterioloăiskā metode, veikta
fermentatīvo īpatnību noteikšana ar komerciālās metodes „BBLTM CrystalTM” palīdzību, ica un
aap gēnu noteikšana ar PCR metodes palīdzību un paraugu vizualizācija ar elektroforēzes
palīdzību. Iegūto rezultātu interpretācijai un apstrādei tika izmantota programma „Statistica
7” (loăiskās regresijas binominālais sadalījums).
Pētījuma aktualitāte slēpjas faktā, ka uz plašu S.aureus izplatības, rezistences
pētījumu fona KONS ir pētīti krietni mazāk. S.epidermidis klīniskā atklāšana izolātā ne
vienmēr Ĝauj izdarīt secinājumus par to, vai mikroorganisms ir infekcijas slimības ierosinātājs,
vai arī tas vienkārši liecina par nespecifisku kultūras inficēšanos. Virulences gēnu izpēte un
noteikšana Ĝaus diferencēt to inficēšanās veidus un ceĜus. Tāpat šis pētījums parādīja aap gēna
vadošo lomu infekcijas procesu izraisīšanā klīniskajos paraugos. Šos rezultātus var izmantot
tālākā nozokomiālo infekciju izplatīšanās izpētē un tālākā koagulāzes negatīvo stafilokoku

This Thesis includes research on Coagulase Negative Staphylococci (CNS), their
phenotypic and genetic characteristics determining the virulence of micro-organisms.
Financial support has been provided as a grant for scientific projects of the Latvian Scientific
Council. The main part of research was performed in the Laboratory for Bioanalytical
Methods at the University of Latvia (supervisor Dr. biol. Jānis Ancāns). Purchase of
equipment was European Union ERAF grant-funded.
The main objectives of Thesis were to isolate genes and enzymes determining
virulence, to compare their presence in the clinical sample group and control group as well as
to determine the link between these genes and the pathogenicity of micro-organisms.
The nature of problem put forward in the Thesis lies in determining the pathogenic
potential of CNS including the most common staphylococcus - S.epidermidis. This type of
staphylococci is a part of normal human skin microflora though in some cases it appears as
nosocomial pathogen. Normally, infections induced by S.epidermidis develop with underlying
lowered immunity and are associated with use of medical equipment. Also, research included
in this Thesis reveals the role of virulence genes ica and aap in the mechanism of biofilm
formation and spread of nosocomial staphylococcus.
Thesis uses the bacteorogical method of sample identification, performs determining
the fermentative properties with commercial method “BBLTM CrystalTM”, determining ica and
aap genes with PCR method as well as visualization of samples, by using electrophoresis. The
interpretation and processing of findings was performed by program “Statistica 7” (binominal
distribution for logistic regression).
Research topicality is specified by fact that CNS are studied much lesser than the
spread and resistance of S.aureus. Clinical revelation of S.epidermidis in the isolate does not
always allow making conclusions whether micro-organism is infection pathogen or a simple
proof of non-specific infection of the culture. Study and determination of virulence genes will
allow differentiate their infection types and ways. This research showed the leading role of
aap gene in inducing the infection process in the clinical samples. These results may be used
for further study of spread of nosocomial infections and study of coagulase negative
staphylococci.
