An ultra-high performance liquid chromatography coupled to high resolution Orbitrap mass spectrometry (UHPLC-Orbitrap-MS) was developed in this master paper and demonstrated as a promising detection technique, applied in the development of the high-throughput method for the routine analysis and control of mycotoxins and pesticide contamination in spices. The elaborated procedure was compared with the conventional triple quadrupole tandem mass spectrometric detection (UHPLC-QqQ-MS/MS). The values of recovery (75-120%) and repeatability (10-15%) for both methods, calculated as the average (n = 5) from the results of spiked (10-500 µg kg-1) paprika samples, showed a good conformity to relevant EU guidelines. In general, the proposed UHPLC-Orbitrap-MS method indicated a sufficient correlation with the method based on the UHPLC-QqQ-MS/MS technique. The developed HRMS method was applied for the analysis of fifty ground paprika samples containing blends of harvested in India and China sweet and hot paprika. The results of analysis demonstrated a presence of two mycotoxins and twelve pesticide residues. The concentrations of determined contaminants were below the MRLs set by the Regulations of the European Union with exception of iprovalicarb violated the EU MRL in two samples. Results of this master paper demonstrated the proposed HRMS method as a promising technique suitable for the assessment of mycotoxins and pesticide residues in complex matrices of dried paprika in accordance to EU legislation.
