Stafilokoku kolonizējošo īpašību izpēte mīkstajās kontaktlēcās

Bakalaura darbs uzrakstīts latviešu valodā uz 33 lapām, satur 2 tabulas, 9 zīmējumus, 26 atsauces uz literatūras avotiem un 3 pielikumus.
Atslēgas vārdi: ādas un acu normālā mikroflora, Staphylococcus, koagulāzes negatīvie stafilokoki (KONS), Stafilokoku ierosinātās acs slimības.
Mērķis: atrast atšķirības starp ādas komensālās mikrofloras adhezīvajiem stafilokokiem un invazīvajiem celmiem, kas kolonizē polimērus.
Uzdevumi: noteikt mīksto kontaktlēcu stafilokoku floras sugu spektru; izveidot no mīkstājām kontaktlēcām izolēto KONS kutūru kolekciju; izdalīt no mīkstajām kontaktlēcām izolēto Staphylococcus epidermidis šūnu DNS un noteikt divus adherences gēnus – ica un aap; iegūtos rezultātus salīdzināt ar nekolonizējošo kultūru DNS analīzes rezultātiem.
Rezultāti: No 30 stafilokoku kultūrām, kas tika izolētas no mīkstajām kontaktlēcām, 4 bija koagulāzes pozitīvie Staphylococcus aureus, visi pārējie bija KONS (koagulāzes negatīvo stafilokoku grupa). Tas norāda, ka KONS ir visbiežāk sastopamie stafilokoki kontaktlēcu mikroflorā.
Izdalītajā KONS kultūru kolekcijā ir 26 Staphylococcus epidermidis celmi , 2 Staphylococcus warneri, 1 Staphylococcus hominis, 1 Staphylococcus capitis. Piecas no visām kultūrām tika izmantotas darbam gēnu noteikšanai, bet pārējās tiek uzglabātas Traumotoloģijas un ortopēdijas slimnīcas mikrobioloģijas labarotorijā.
No 5 Staphylococcus epidermidis kultūrām tika izolēta DNS un ar polimerāzes ķēdes reakciju (PCR) noteikti divi adherences gēni – ica un aap. Ica gēni netika konstotēti izolētiem paraugiem, bet aap gēni bija pozitīvi vienā no pieciem paraugiem. Iegūtie rezultāti ir ticami, jo pozitīvs rezultāts bija pozitīvai kontrolei, kura uzrāda, ka PCR reakcija ir notikusi.
Salīdzinot ar gēnu karti, kas iegūta no deguna gļotādas normālās mikrofloras stafilokokiem, būtiskas atšķirības nav konstatētas. The Bachelor Work is written in Latvian and it contains 33 pages, 2 tables, 9 pictures, 26 literature references and 3 appendices.

The aim: To find differences between Adhesive Staphylococcus of skin commensally micro flora and invasive stems which colonize polymers?
Objectives: To identify the spectrum of species of Staphylococcus flora on soft contact lens; to create the collection of CONS culture, isolated from soft contact lens; to isolate the DNA of Staphylococcus epidermidis cell, taken from a soft contact lens, and identify two adherence genes – ica and aap; to compare the findings with the results of the analysis of non-colonizing DNA cultures.
Results: from 30 cultures of Staphylococcus that were isolated from a soft contact lens, 4 were coagulase-positive Staphylococcus aureus, the rest were CONS (a group of coagulase-negative Staphylococcus) showing that CONS are the most widespread Staphylococcus in the micro flora of contact lenses.
There are 26 stems of S. epidermidis, 2 S.warneri, 1 S.hominis, 1 S.capitis in the isolated collection of KONS cultures. Five cultures of fifteen were used for the further identification study of genes but the rest are being kept in the laboratory of microbiology in the Hospital of Traumatology and Orthopaedy.
The DNA was isolated from 5 cultures of S.epidermidis and two adherence genes – ica and aap were identified by the polymerase chain reaction. Ica genes were not found in isolated samples but aap genes were positive of one in five samples. The obtained findings are valid because a positive control, showing that PCR reaction took place, had a positive result.
Comparing with a gene card, taken from the micro flora of Staphylococcus in the nasal mucous membrane, no essential differences were found.
